封闭接头配套试剂
产品介绍
在杂交捕获过程中,接头之间的退火杂交,会在不同插入片段的文库之间形成串联结构,探针在捕获目标区域片段的同时也会捕获与之串联的非靶标片段,导致On-Target低,有效数据减少。而特异性1对1 Blocker使用较为繁琐,容易出现操作失误。因此,我们为MGI和Illumina测序平台均开发了相应的Universal Blocker,可高效封闭不同Index文库,同时兼容单端和双端Index等多种文库结构。
以Universal Blockers-ILMN-TS(Du)为例说明,该产品是针对illumina Truseq接头开发的通用阻断剂,如图所示,可适用于单端、双端以及双端+UMI等Truseq接头文库。

有效避免标签错配
多文库捕获时,阻断剂设计缺陷,合成质量低都可能造成index-hopping现象,导致样本之间的污染。使用伯科Universal Blockers-ILMN-TS(Du)和友商I产品对已知SNP的两个样本进行2-Plex杂交捕获,通过分析等位基因频率对index-hopping现象进行检测。
